Initiator caspases

WebbThe caspase family of proteases is central to this apoptotic network, with initiator and executioner caspases, and their interaction partners, regulating and executing apoptosis. In this work, we interrogate and modulate this network by exogenously introducing specific initiator or executioner caspase proteins. WebbCaspase 7 is an executioner caspase that was identified based on its homology with caspases 1 and 3, as well as the C. elegans cell death protein CED-3. Alternative splicing of Caspase-7 mRNA results in the production of 3 distinct isoforms. Caspase-7 activity can be directly inhibited by XIAP expression.

Apoptosome: a platform for the activation of initiator caspases

WebbCaspases are central components of the machinery through cleavage at specific internal Asp residues that responsible for apoptosis. Recent structural and bio-separate the large and small subunits (Figure 1). Webbv-FLIPs are viral proteins that were first identified as modulators of apoptosis; they contain two death effector domains, which are also found in some initiator caspases such as procaspase-8. These v-FLIP proteins can be recruited to the DISC through the binding of the death effector domain to similar domains in the adaptor proteins, but are otherwise … Webb13 apr. 2024 · Caspases are essentially the executioners of apoptosis. Webb19 aug. 2005 · Caspases are responsible for the execution of programmed cell death (apoptosis) and must undergo proteolytic activation, in response to apoptotic stimuli, to function. The mechanism of initiator caspase activation has been generalized by the induced proximity model, which is thought to drive dimerization-mediated activation of … Webb31 okt. 2024 · CTSB, either directly or indirectly, plays a critical role in activating the apoptotic pathway via initiator caspases rather than executioner caspases [ 18 ]. Indirect regulation of caspases by CTSB is mediated through mitochondrial membrane degradation, which translocates apoptosis-inducing components from the mitochondria …

As is the case for most proteases, caspases are expressed in cells as inactive zymogens also known as procaspases (Figure 1a).All mature caspases form a head-to-tail dimer with each unit composed of a large and small subunit containing one active site, as illustrated by the crystal structure of caspase-7 (Figure 1b).Some …

Webb5 juni 2024 · Caspases are members of the cysteine-1 aspartate-specific protease family [ 15, 16 ]. There are major groups of caspases in mammals, including initiator caspases and executioner caspases. Caspases 2, 8, 9, 10, 11 and 12 are known to be initiators, and caspases 3, 6 and 7 are known to be executioners [ 17, 18 ]. Webb1 dec. 2002 · Initiator caspases may directly process and activate executioner caspases 3, 6, and 7 (for review, see Hengartner, 2000). Caspases can selectively be inhibited by small peptides, mimicking the substrate recognition site, carrying electrophiles such as aldehydes, nitriles, or ketones at their C terminus that react with the active site Cys.
